A seminar by Dr Asokan Chinnasamy highlights the role of blood grouping Hybridoma cell line on increasing the secretion of monoclonal anitobodies (mAbs) for diagnostic/therapeutic purposes.
Dr Chinnasamy, from the School of Medical Sciences, gave a presentation on his paper to the UKH staff.
Biopolymer membrane was
prepared using two oppositely charged natural polymers. The biopolymer membrane
was used for the encapsulation of Hybridoma Anti-D cell line to produce
monoclonal antibodies (mAbs). Soy Hydrolysate is one of the supplement and replacement
of Fetal calf serum for the growth of the mammalian cells and secretion of the
biological products. It contains low molecular weight nutrients, peptides and
free amino acids which are essential for cell growth.
beads and the cell growth were observed in inverted Microscope. Soy Hydrolysate
was used as animal component free supplement with the different concentrations
of 0.1%, 0.3%, 0.5% and 1% with RPMI 1640 medium to enhance the cell growth and
increase the monoclonal antibodies secretion. The effect of Soy Hydrolysate is
studied by the cell growth and monoclonal antibodies secretion. The Increased
secretion of monoclonal antibodies is confirmed by Avidity test, Titer
analysis, total protein estimation and size exclusion chromatography analysis.
The present study concludes
that increase the yield of diagnosis/therapeutic monoclonal antibodies could be
maximized by the encapsulation method. Thus, in the optimization of monoclonal
antibodies in cell culture and their increased secretion, additives like Soy
Hydrolysate plays a significant role.
